The match is scheduled to start at 7pm local time (2pm GMT) on Wednesday, February 15.
The Multan Cricket Stadium in Multan will host this fixture.
Last season's runners-up, Multan Sultans began their new campaign with a loss by one run to Lahore Qalandars in the opening game of the new campaign.
The Gladiators are hoping they can turn their form around against the Sultans, having not beaten them since March 2021. They finished last season in fifth place with four wins and six losses.
Mohammad Rizwan and opening partner Shan Masood made an impressive start in chase of 176 runs in their first game of the season. The skipper made 75 runs from 50 deliveries, with Masood chipping in with 35. Sadly, the rest of the batting order could not finish the job started by the opening pair.
England's Jason Roy and Will Smeed could open the batting for the Gladiators and form a destructive partnership. The likes of Martin Guptill and Iftikhar Ahmed are also available meaning the line-up is not lacking in firepower. Nineteen-year-old right-arm fast bowler Naseem Shah will look to be a huge part of the Gladiators' attack.

Rilee Roussow did not play in Multans Sultans' opening game after the delayed SA20 final; however, he could look to make his return for this game. Shahnawaz Dahani has been ruled out for the rest of the season with a broken finger. Mohammad Ilyas has been drafted as a replacement by Multan.
Mohammad Hafeez has replaced the injured Ahsaan Ali in the Quetta Gladiators' squad.
